Rybelsus is the only oral form of semaglutide — the same active molecule in the injectable medications Ozempic and Wegovy. FDA-approved in 2019 for type 2 diabetes management, it gives patients who are unwilling or unable to self-inject access to the proven weight loss and metabolic benefits of semaglutide without needles.
Oral semaglutide uses a proprietary absorption enhancer called SNAC (sodium N-(8-[2-hydroxybenzoyl] amino) caprylate) that allows semaglutide to survive the gastric acid environment and be absorbed through the stomach lining. Because of this mechanism, Rybelsus must be taken on an empty stomach with no more than 4oz (half a glass) of plain water, and you must wait at least 30 minutes before eating, drinking, or taking other medications. This is critical for absorption — food significantly reduces bioavailability.
While oral semaglutide produces somewhat less weight loss than injectable semaglutide at equivalent doses (due to variable oral absorption), it remains significantly effective. In clinical trials, patients on 14mg oral semaglutide lost approximately 4–6 kg over 26 weeks, with greater results seen in longer-duration studies and when combined with dietary changes.
